Cytokines
Also called: inflammatory cytokines, bone cytokines
What it means
Cytokines are signaling proteins from immune cells; some, like TNF-alpha, IL-1 and IL-6, promote inflammation and can speed up bone breakdown when chronically elevated.
Ongoing inflammation from these cytokines is a real driver of bone loss, which is why inflammatory conditions and low estrogen both raise fracture risk.
Cytokines are small signaling proteins your immune cells release to communicate with each other and with other tissues, including bone. Some cytokines, especially TNF-alpha, IL-1 and IL-6, are described as pro-inflammatory because they ramp up inflammation. In bone, their main trick is boosting RANKL signaling, which switches on more osteoclast activity — the cells that break down bone. More resorption without matching new bone formation tips the balance toward bone loss over time.
This is why ongoing inflammatory conditions like rheumatoid arthritis and inflammatory bowel disease are recognized risk factors for lower bone density. It's also part of why estrogen matters so much: estrogen normally helps keep these inflammatory cytokines in check and supports osteoprotegerin (OPG), a natural brake on RANKL. When estrogen drops at menopause, that brake loosens and cytokine-driven bone turnover can pick up.
It's worth being careful here: 'leaky gut' is a real, measurable phenomenon in specific diagnosed conditions, but it isn't something a home test can confirm, and commercial protocols claiming to fix it lack solid outcome evidence. If you have an inflammatory condition and are concerned about your bones, a conversation with your doctor about monitoring and appropriate testing is the most useful next step.
How sure is this?
Graded Emerging Evidence: Promising research, but findings are still developing and may be primarily from laboratory or animal studies. Read each study with its grade in mind. One study is a piece of the picture, not the whole of it.
Chronically elevated inflammatory cytokines like TNF-alpha and IL-6 drive bone breakdown and raise fracture risk.
Where this leads
WordYou just read that cytokines can speed bone breakdown. Curious how one specific cytokine, TNF-alpha, quietly drains bone strength?
