LRP5/LRP6
Also called: LRP5 gene, LRP6 receptor, Wnt co-receptor
What it means
LRP5 and LRP6 are receptors on bone cells that work with the Wnt signaling pathway to control how much new bone your body builds.
This pathway is one of the body's main switches for building bone, and it's the target behind some modern osteoporosis treatments.
LRP5 and LRP6 are proteins that sit on the surface of bone-building cells and act as co-receptors for the Wnt signaling pathway. Think of them as docking stations: when Wnt proteins bind to LRP5/LRP6 together with another receptor, they trigger a chain of signals inside the osteoblast that tells it to keep building bone instead of slowing down.
This pathway matters because it helps set how much bone you make over your lifetime. Genetic variations in LRP5 are a classic example used in research to show how strongly bone formation can be turned up or down — some rare mutations lead to unusually high bone density, while others are linked to very low bone density and fragile bones.
LRP5/LRP6 signaling is also the target of a natural brake called sclerostin, made by osteocytes, and a related inhibitor called Dickkopf-1. When these blockers attach to LRP5/LRP6, they quiet the Wnt pathway and reduce new bone formation. Some osteoporosis medications are designed around this exact biology, and your doctor can explain how a specific treatment fits your situation.
LRP5 and LRP6 receptors work with the Wnt pathway to control new bone building and are a target for modern treatments.
Where this leads
WordLRP5/LRP6 work with the Wnt pathway to control bone building. Curious how this whole signaling system switches bone-building cells on or off?
